Gold nanoshells aided optical imaging in vitro, in vivo, and ex vivo

چکیده
Scatter-based optical imaging technologies such as Optical Coherence Tomography (OCT) and Reflectance Confocal Microscopy (RCM) offer a unique non-invasive approach to the detection, diagnosis, and monitoring of cancer. In this article, we describe the use of targeted NIRtuned gold nanoshell for imaging in vitro, ex vivo and in vivo. Nanoshells are a new class of optically active nanoparticles with tunable plasmon resonances based on geometric construction. These particles can be easily tuned to absorb or scatter strongly within the wavelengths of 6501300 nm, known as the near infrared region. This region is of significant biological importance providing a therapeutic window and imaging applications in tissue as the primary components of tissue, blood and water, do not have significant absorption coefficients in this range of wavelengths.

Photothermal optical coherence tomography in ex vivo human breast tissues using gold nanoshells.
We demonstrate photothermal optical coherence tomography (OCT) imaging in highly scattering human breast tissue ex vivo. A 120 kHz axial scan rate, swept-source phase-sensitive OCT system at 1300 nm was used to detect phase changes induced by 830 nm photothermal excitation of gold nanoshells. Localized phase modulation was observed 300-600 microm deep in scattering tissue using an excitation po...
متن کاملRapid Stereomicroscopic Imaging of HER2 Overexpression in Ex Vivo Breast Tissue Using Topically Applied Silica-Based Gold Nanoshells
Tumor margin detection for patients undergoing breast conservation surgery primarily occurs postoperatively. Previously, we demonstrated that gold nanoshells rapidly enhance contrast of HER2 overexpression in ex vivo tissue sections. Our ultimate objective, however, is to discern HER2 overexpressing tissue from normal tissue in whole, nonsectioned, specimens to facilitate rapid diagnoses. Here,...

Gold nanoshells (GNS) are a new class of nanoparticles that can be optically tuned to scatter or absorb light from the near-ultraviolet to near-infrared (NIR) region by varying the core (dielectric silica)/shell (gold) ratio. In addition to spectral tunability, GNS are inert and bioconjugatable, making them potential labels for in vivo imaging and therapy of tumors. We report the use of GNS as ...
متن کاملApplications of gold nanoparticles in cancer nanotechnology.
It has been almost 4 decades since the "war on cancer" was declared. It is now generally believed that personalized medicine is the future for cancer patient management. Possessing unprecedented potential for early detection, accurate diagnosis, and personalized treatment of cancer, nanoparticles have been extensively studied over the last decade. In this review, we will summarize the current s...
متن کاملLinarin Inhibits the Acetylcholinesterase Activity In-vitro and Ex-vivo
Linarin is a flavone glycoside in the plants Flos chrysanthemi indici, Buddleja officinalis, Cirsium setosum, Mentha arvensis and Buddleja davidii, and has been reported to possess analgesic, antipyretic, anti-inflammatory and neuroprotective activities. In this paper, linarin was investigated for its AChE inhibitory potential both in vitro and ex vivo. Ellman’s colorimetric method was used for...
